HighYield T7 Biotin11 RNA Labeling Kit (CTP-based) is designed to produce randomly Biotin-modified RNA probes with a 11-atom linker via in vitro transcription. Such probes are ideally suited for in situ hybridization and Northern Blot experiments. \nBiotin-11-CTP is efficiently incorporated into RNA as substitute for its natural counterpart CTP using an optimized reaction buffer and T7 RNA Labeling Polymerase Mix.\n35 % Biotin-11-CTP substitution typically results in an optimal balance between reaction and labeling efficiency. Individual optimization of Biotin-11-CTP/CTP ratio however, can easily be achieved with the single nucleotide format. \nThe resulting Biotin-modified RNA probe with a 11-atom linker can subsequently be detected by HRP- or AP-modified streptavidin. \nThe kit contains sufficient reagents for 30 labeling reactions of 20 μl each (35 % Biotin-11-CTP substitution, 1 mM ATP, GTP, UTP, 0.65 mM CTP, 0.35 mM Biotin-11-CTP).
