    Flow-cytometry using anti-GITR antibody YGITR860.103.5. Mouse spleenocyts were stained with an isotype control (3.0 - Orange) or the rmeric version of YGITR860.103.5 (orb1672029) at a concentration of 1 ug/ml for 30 mins at RT without (Blue) and with (Grey) stimulation of the leukocytes with immobilized anti-CD3 and anti-CD28 antibodies. After washing- bound antibody was detected using a AF488 conjugated donkey anti-ribody and cells analysed on a FACSCanto flow-cytometer.

    Immunofluorescence staining of fixed mouse splenocytes with anti-GITR (Tumor necrosis factor ligand superfamily member 18) antibody YGITR 860.103.5. Immunofluorescence analysis of paraformaldehyde fixed mouse (Mus musculus) splenocytes on Shi-fix™ coverslips- permewith 0.15% Triton stained with the chimeric rsion of YGITR 860.103.5 (orb1672029) at 10 ug/ml for 1h followed by Alexa Fluor® 488 secondary antibody (1 ug/ml)- showing membrane and some cytoplasmic staining in a subset of cells. The nuclear stain is DAPI (blue). Panels show from left-right- top-bottom orb1672029- DAPI- merged channels and an isotype control. The isotype control was stained with an anti-Fluorescein antibody (0) followed by Alexa Fluor® 488 secondary antibody.
