Description | Epstein-Barr virus (EBV) is a prevalent gamma herpesvirus, infecting upwards of 90% of the world population. EBV is associated with several cancers, including nasopharyngeal carcinoma, Burkitt’s lymphoma and Hodgkin’s lymphoma. EBV nuclear antigen-1 (EBNA1), the first reported EBV latency protein, is expressed in all EBV-associated tumors and all latency programs of the virus. It constitutes a marker that discriminates the virus-associated cancer cells and normal cells, and thereby offers opportunities for targeted therapeutic intervention.EBNA1 plays a vital role in viral DNA replication and the episomal genome maintenance through sequence-specific DNA binding. Furthermore, EBNA1 interacts with viral and cellular promoters and thereby conduces to the transcriptional regulation of both viral and cellular genes. Several evidences demonstrate that EBNA1 contributes to the development of EBV-associated tumors via increasing cell proliferation and survival and possibly by inducing oxidative stress. |
