Measured by its binding ability in a indirect ELISA. Immobilized Human AFP protein, His Tag at 2 µg/mL (100 µl/well) can bind Mouse Anti-Human AFP Antibody, the EC50 is 24.94 ng/ml.

25 ug total protein per lane of various lysates probed with AFP (A2) monoclonal antibody, unconjugated (orb4247) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r.t. for 60 min.

Paraformaldehyde-fixed, paraffin embedded (Rat liver), Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min, Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes, Blocking buffer (normal goat serum) at 37°C for 30 min, Antibody incubation with (AFP (A2)) Polyclonal Antibody, Unconjugated (orb4247) at 1:400 overnight at 4°C, followed by a conjugated secondary antibody (orb868589) for 90 minutes, and DAPI for nuclei staining.
