1. CD3: Cluster of Differentiation 3

The CD3 T-cell co-receptor critically assists in the activation of both cytotoxic T-cells (CD8+ naive T-cells) and T helper cells (CD4+ naive T-cells).

Present natively as a protein complex, it is strictly comprised of four distinct chains. In mammals, this functional complex contains a gamma (γ), a delta (δ), and two identical epsilon (ε) chains. The CD3γ, CD3δ, and CD3ε chains are highly related cell-surface proteins that each contain a single extracellular immunoglobulin domain.

A Universal T-Cell Marker

The CD3 antigen can be found exclusively bound to the membranes of all mature T-cells and is functionally present in virtually no other cellular lineage. This extreme cellular specificity, actively combined with its persistent presence at all stages of T-cell development, makes it the absolute gold-standard, universal T-cell marker for IHC tissue sectioning and Flow Cytometry. The antigen is also highly persistent in almost all T-cell lymphomas and leukaemias, robustly allowing for their immediate clinical differentiation from similar B-cell and myeloid neoplasms.
